Background
SIRT1 is a nuclear protein and member of the sirtuin protein family of deacetylating proteins.  SIRT1 is involved in regulating a number of cellular pathways including apoptosis and cell senescence, longevity and aging (1-3).  Not surprisingly, SIRT1 targets to forkhead (FOXO) transcription factors.  SIRT1 physically binds to the p65/RelA protein in the NF-kB complex and inhibits the transcriptional activity by deacetylating the p65 component (4).  SIRT1 anti-inflammatory responses have also been demonstrated  using resveratrol, a potent activator of SIRT1, which has been shown to potentiate the SIRT1-mediated anti-inflammatory response (5).  Detailed involvement of SIRT1 and NF-kB, and the control and interconnection of the Innate Immune and inflammatory response and aging, is reviewed by Salminen et al (6).

Antigen
A portion of amino acids 400-450 from human SIRT1 was used as the immunogen for this antibody.

Application Notes
The amino acid sequence used as immunogen for this antibody is 100% homologous in human, mouse, rat, orangutan, chimpanzee, panda, carolina anole, cow, new world monkey, dog, chinese hamster, horse, chicken, mole rat, rhesus monkey, wild turkey, opossum, platypus, rabbit, pig, zebra finch, frog, cat, and white-cheeked gibbon, and 89% homologous in zebrafish, cichlid fish, and killifish.

SIRT1 Antibody

Western blot analysis of SIRT1 using SIRT1 antibody. HeLa lysate probed with 1) prebleed, 2) SIRT1 antibody and 3) mouse NIH 3T3 lysate probed with SIRT1 antibody at 2 ug/ml. IMGENEX’s goat anti-rabbit Ig HRP secondary antibody (20301) and PicoTect ECL substrate solution (10087K) were used for this test.
